Pantoprazole

Pantoprazole is a proton pump inhibitor that inhibits the Hydrogen Potassium (H+/K+) pump present in the parietal cells of the stomach. It thus inhibits the gastric secretion of acid.

Acid secretion is inhibited after 15 – 30 minutes of intravenous administration and 2.5 hours of oral administration. The maximum effect can be seen 2 hours after intravenous administration.

Oral pantoprazole is rapidly absorbed and the duration of action lasts for 24 hours. 98% of the drug is bound to proteins, primarily to albumin.

It has a bioavailability of 77%. The half-life elimination in neonates is about 3 hours, children after intravenous and oral administration are 1.22 and 1.27, and adults is one hour.

The half-life elimination is increased to 3.5 – 10 hours in patients with CYP2C19 deficiency.

It is excreted primarily in the urine (71% as metabolites) and feces (18%). The clearance of the drug also increases with age and the increasing weight of the individual.
